Curcumin in Anti-Inflammatory and Antioxidant Studies

Curcumin is known as a powerful anti-inflammatory. Many people in the world today take it to control pain by arthritis, fibromyalgia, and other painful conditions. A primary reason for this is that Curcumin does not have the severe side effects that many over-the-counter pain remedies have such as liver damage from acetaminophen. If anything, when it concerns the liver, Curcumin is popular for its detoxifying effects.

In case study for a brand name of Curcumin called Theracumin, 50 patients aged 40 years old and over with knee osteoarthritis of Kellgren-Lawrence grade II or III were enrolled in a randomized, double-blind, placebo-controlled, prospective clinical study, 180mg of Theracumin or a placebo was given to the patient orally every day for eight week. Monitoring was done by blood biochemistry analysis before and after a week of each intervention. Symptoms were rated at zero, two, four, six, and eight weeks with the Japanese Knee Osteoarthritis Measure, knee pain visual analog scale (VAS), knee scoring system of the Japanese Orthopedic Association, and the need (dependency) for non-steroidal anti-inflammatory drugs. After eight weeks of treatment, knee pain VAS scores were significantly lower in the Theracumin group versus the placebo group, and dependence on celecoxib was considerably lower than the placebo group.  In a nutshell, what this study proves is that this particular Curcumin compound not only reduced pain but reduce pain but reduced the dependence patients had on non-steroidal anti-inflammatory drugs, most likely because of its detoxifying effects on the liver.

In this study, nanoemulsion Curcumin (NEC) was demonstrated to be an effective in-vitro immunosuppressive in an individual mouse model containing the reporter gene NF-kB-luciferase.

The nanoemulsion Curcumin treatment seemed to inhibit the secretion of monocyte chemoattractant protein-1, also known as MCP-1. Futhermore, the expression of toll-like receptor 4 (TLR4) and the RA gene were both observed to have decreased levels. Lipopolysaccharide (LPS) induced phosphor regulation was also blocked in the subunit P-65 of the nuclear factor kappa-light-chain-enhancer of activated B-cells (NF-kB) protein complex, and in murine macrophages’Ba.

In pericarditis presented in a mouse model, nanoemulsion of Curcumin was ineffective at the reduction of B and T cells levels. However, NEC was able to reduce the recruitment of macrophages significantly. Cell migration was also significantly inhibited by the treatment of primary human macrophage monocyte-derived cell lines with Curcumin, in vitro. These results helped to show that inflammation suppression can be accomplished via Curcumin treatments that successfully reduce macrophage migration through multiubiquitin-chain-binding protein (Mcb1) and (NFkB) protein complex inhibition. The data gathered from this study could be pivotal in establishing nanoemulsion formulas as a bioavailable form of Curcumin useful for medical treatment. This particular technology did increase bioavailability for absorption in the human body, and the formulation caused the suppression of protein and chemical factors known to cause inflammation within the cell.

Laboratory research has profoundly demonstrated anti-inflammatory, antioxidant, and anticancer properties of Curcumin. Curcumin inhibits the properties of amyloid  B peptides, AB, and Curcumin inhibits the expression of AB-induced Egr-1 and Egr-1 DNA binding activity. It also prevents TNF (tumor necrosis factor), alpha and interleukin-1 beta, cyclooxygenase or COX-2, phospholipases, the transcription factor and enzymes involved within metabolizing membrane phospholipids into prostaglandins. Curcumin decreases and prohibits the main chemicals for inflammation and decreases the transcription of inflammatory cytokines, as well as hindering  AP-1 and NF-Kappa B.  People who are overweight and those who cannot lose weight may find that it is due to toxins present in their environment, food sources, stress, medication and illness. Cells suffer from inflammation because they in cells use it as a defense mechanism when they feel threatened or injured, hence not functioning as usual. Toxins usually get stored in lipid, adipose, or fat cells, and this creates a problem with the metabolism not being fully functional, resulting in cells becoming radical and inflamed. Curcumin, a great liver detoxifier,  delivers antioxidants to the cell and regulates protein function to keep unwanted inflammatory agents out. It helps the liver strip the toxins from the body to make it operate in a healthier manner, thus helping in weight loss.

A ship lands and sailors (curcumin, antioxidants, amino acids, vitamins, and nutrition) get off the boat.  The sailors are a new crop of boyfriends. Each angry girl gets a new boyfriend, making her content and happy, restoring tranquility to the town. Sailors eventually leave the city, but the town has taken measures to ensure sailors keep landing (proper care and nutrition of the body).  

That is what nutrients, amino acids, and vitamins do for the cell.  They take the unpaired electron and pair up with it within the molecules, which is atoms, or a combination of molecules with sets of paired electrons. When electrons are unpaired, they are like hot pokers bouncing around the cell, causing damage which eventually causes death. If there is no intervention from beneficial amino acids, nutrients, or vitamins, they cause inflammation, which is usually the first sign of disease or dysfunction within the body,
